Description: “Standards and Guidelines for the Interpretation and Reporting of Sequence Variants in Cancer”, developed by AMP with representation from American College of Medical Genetics and Genomics, American Society of Clinical Oncology (ASCO), and College of American Pathologists (CAP), was published in 2017 as an AMP/ASCO/CAP joint consensus recommendation. This guideline established a four-tier system for classifying somatic variants that has been widely adopted in clinical care. This session will discuss the multidisciplinary collaborative AMP Clinical Practice Committee Working Group's upcoming guideline and the development of new and updated recommendations, reflecting current practices and technological advancements in cancer genomics.
Learning Objectives:
- Review the tiered system for somatic variant classification and its relationship with the five levels of evidence of clinical significance, including oncogenicity assessment and clinical assessment.
- Discuss updated recommendations for the evaluation and reporting of germline pathogenic/likely pathogenic variants identified during tumor sequencing.
- Discuss additions and clarifications within the upcoming guideline, including reporting of hematologic oncology-related variants, mutational signatures, and other complex biomarkers.
